SpiroChem’s macrocycle libraries are meticulously designed to encompass a wide range of chemotypes while maintaining strategic diversity in key molecular parameters. Depending on your project needs, SpiroChem offers ready-to-screen collections or custom-designed libraries, leveraging extensive expertise in macrocycle synthesis.
The SpiroChem Macrocycle Library compounds are based on a semi-peptidic design concept. All compounds are purified and analytically characterized.
In contrast to the competitive landscape, the library is CHEMICALLY ENABLED: rapid follow-up chemistry is ensured by a highly sophisticated internal macrocycle chemistry platform (Hercules+ platform) based on solid-phase chemistry that underwent a 10-year optimization phase.
From that library, numerous successful drug discovery projects emerged throughout the international pharma and biotech industry, exemplified by two recent publications from Roche, where a novel clinical candidate, Zosurabalpin (RG6006) was described as an inhibitor of a bacterial lipopolysaccharide transporter (https://doi.org/10.1038/s41586-023-06799-7 and https://doi.org/10.1038/s41586-023-06873-0). The original hit was identified from an earlier version of the SpiroChem’s library and optimized based on the well-established chemistry concepts around the macrocycle collection.
Two standard formats are offered:
The libraries are available in DMSO solution, ready for screening. Volume, concentration, plate reference and layout are customizable, ensuring seamless integration into your screening workflows.
The apparent complexity of the structures is decorrelated from their synthetic accessibility. The chemical space surrounding the macrocycle libraries is fully enabled thanks to our proprietary Hercules+ platform: we guarantee a rapid turn-around time for hit validation and downstream optimization work.
