Spirocycles


Spirocycles are rigid scaffolds that have attracted the attention of medicinal chemists over the last decade. They are three-dimensional fragments and have a high sp3/sp2 ratio by nature, a feature that is of increasing importance in the design of new classes of biologically-active molecules with improved properties. They also offer conformational restriction in the positioning of exit vectors.


Why Spirocycles?


Spirocycles are rigid scaffolds that have attracted the attention of medicinal chemists over the last decade. They are three-dimensional fragments and have a high sp3/sp2 ratio by nature, a feature that is of increasing importance in the design of new classes of biologically-active molecules with improved properties. They also offer conformational-restriction in the positioning of exit vectors. Two intrinsic properties of spirocycles are worth mentioning:1 1) they have a reduced lipophilicity compared to analogous cyclic molecules and 2) they offer many more opportunities for covering the chemical space and positioning exit vectors, much more three-dimensional character.

Researchers in the Carreira group at ETH-Zürich and at Roche, Basel, as well as other academic and industrial researchers, have demonstrated that the use of spirocycles in drug discovery often leads to improved physicochemical and improved pharmacokinetic properties as well as structural novelty. Their rigidity makes them ideal scaffolds to orientate exit vectors in a conformationaly-restricted manner.

Another advantage of the Spirocycles is that their introduction in molecules of interest usually comes with novel chemical space and therefore new intellectual property.
