Our bicyclo[x.y.z]alkanes series are rigid carbocycles with one or two exit vectors positioned at the bridge-head positions. They can serve as rigid linkers, spacers or scaffolds and offer a great diversity for the orientation of exit vectors in a rigid manner.

Why Bicyclo[x.y.z]alkanes?

Our bicyclo[x.y.z]alkanes series is a useful addition to our portfolio of innovative chemical tools designed in a continuous effort to provide as many solutions as possible to the medicinal chemistry community for accelerated drug discovery.

With the growing success of the bicyclo[1.1.1]pentanes series, we wanted to extend the chemical space to their homologated versions. For example, the bicyclo[2.2.2]octanes are elongated versions of the [1.1.1] series and offer a linear positioning of exit vectors. Introduction of a bridge of a different length will generate an angle between the two exit vectors as seen in the [2.1.1], [2.2.1], [3.1.1] and [3.2.2] series (Figure), thereby potentially offering a 3-dimensional alternative to arenes with meta substitution patterns.

Our bicyclo[x.y.z]alkanes can also be used as conformationally-restricted analogs of more standard carbocycles (cyclobutanes, cyclopentanes, cyclohexanes), thereby offering a more rigid scaffold, positioning exit vectors in a fixed orientation and reducing the entropy.
